Abstract:
Emerging evidence indicates that the risk of Alzheimer’s disease in individuals who carry the APOE4 allele is modifiable. In this talk, Dr. Yassine argues that everyone should get an APOE genotype and APOE4 carriers need to start a dementia prevention lifestyle as early as possible. He provides examples of how beneficial effects of increased polyunsaturated fat consumption (Mediterranean diet, fish consumption or long-term DHA supplementation) on cognition are stronger in younger and middle aged APOE4 carriers, and how the efficacy of these interventions wane with advancing age and cognitive disease. Similarly, ketogenic diets do not appear to have a beneficial effect on cognitive performance in older APOE4 carriers compared to non-carriers. Mechanisms of reduced nutrient uptake in the aging APOE4 brain are discussed with insights into why future precision medicine will combine drugs and nutritional interventions for dementia prevention.
